Overview

C-10386 is a potent, selective, and orally bioavailable molecular glue degrader (MGD) that targets the translation termination factor GSPT1. Developed by Cyrus Therapeutics, the compound functions by hijacking the Cereblon (CRBN) E3 ligase to induce the rapid, potent, and durable degradation of GSPT1. This degradation leads to dysregulated protein translation termination and subsequent induction of TP53-independent apoptosis, particularly in cells with translational addiction. C-10386 has demonstrated significant anti-tumor activity in pre-clinical models of acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and various MYC-driven solid tumors. In ex-vivo assays using patient-derived primary AML cells, C-10386 showed high sensitivity with nanomolar EC50 values.
