C-CAR-011 is a CD19-targeted chimeric antigen receptor (CAR) T cell therapy developed for the treatment of relapsed or refractory B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL), non-Hodgkin lymphoma, and acute lymphoblastic leukemia. The therapy involves genetically modifying a patient's own T cells to express a CAR that recognizes the CD19 antigen on malignant B cells. This enables the engineered T cells to identify and destroy cancerous B cells. The construct used in C-CAR-011 includes a second-generation anti-CD19 CAR with a 4-1BB costimulatory domain to enhance efficacy and persistence of the modified T cells. Clinical studies have shown promising safety and efficacy results in heavily pretreated patients with refractory DLBCL, with manageable cytokine release syndrome and no observed neurotoxicity[1][2].
