c-FLIP siRNA is an experimental small interfering RNA (siRNA) designed to silence the expression of Cellular FLICE-like Inhibitory Protein (c-FLIP), a key regulator of the extrinsic apoptotic pathway. c-FLIP acts as a catalytically inactive homolog of caspase-8 and caspase-10, competing for binding at the death-inducing signaling complex (DISC) and thereby preventing pro-apoptotic signaling. By downregulating c-FLIP, this siRNA restores sensitivity to apoptosis-inducing ligands like TRAIL and enhances the cytotoxic effects of various chemotherapeutic agents. It has been extensively studied in preclinical models of various malignancies, including cervical, breast, and prostate cancers, to overcome resistance to programmed cell death. It is primarily used as a research reagent and is not currently associated with a single commercial drug development program.
