Overview

c-Jun-overexpressing CD33 CAR T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy designed to treat relapsed or refractory acute myeloid leukemia (AML). The therapy consists of T cells engineered to express a CAR targeting the CD33 antigen, which is highly expressed on AML blasts and leukemic stem cells. A distinguishing feature of this product is the intentional overexpression of the c-Jun transcription factor (a component of the AP-1 complex). This modification is intended to overcome the hyporesponsiveness and impaired signaling (specifically in the ERK and calcium pathways) typically observed in CAR T cells exposed to the AML microenvironment. By overexpressing c-Jun, the cells exhibit enhanced activation, increased production of cytokines like IL-2 and IFN-γ, and upregulated costimulatory molecules, thereby improving anti-tumor efficacy and persistence in myeloid malignancies.
