C-Scape

C-Scape is an investigational oral antibiotic combination consisting of ceftibuten, a third-generation cephalosporin, and clavulanate (clavulanic acid), a beta-lactamase inhibitor. Developed by Achaogen to address the need for oral treatment options for multi-drug resistant (MDR) Gram-negative infections, it specifically targets pathogens producing extended-spectrum beta-lactamases (ESBLs). The combination works by utilizing ceftibuten to inhibit bacterial cell wall synthesis while clavulanate protects the cephalosporin from enzymatic degradation by inhibiting Class A beta-lactamases. The program received significant funding from the Biomedical Advanced Research and Development Authority (BARDA) and completed Phase 1 clinical testing. However, development was halted following Achaogen's Chapter 11 bankruptcy in 2019, and the rights to the candidate were subsequently acquired by Heritage Pharmaceuticals.
