C-SLN + antisense oligonucleotide targeting IGFL2-AS1 is an experimental nanomedicine developed for the treatment of sunitinib-resistant clear cell renal cell carcinoma (ccRCC). The therapeutic consists of an antisense oligonucleotide (ASO) encapsulated within chitosan-modified solid lipid nanoparticles (C-SLN) for targeted delivery. The ASO is designed to silence the long non-coding RNA (lncRNA) IGFL2-AS1, which is significantly upregulated in resistant RCC cells. IGFL2-AS1 normally promotes sunitinib resistance by binding to the RNA-binding protein hnRNPC, which prevents the suppression of TP53INP2, leading to increased autophagy and cell survival. By degrading IGFL2-AS1, the C-SLN/ASO complex suppresses this autophagic pathway and restores the sensitivity of cancer cells to sunitinib. Preclinical evidence in patient-derived xenograft models has shown that intravenous administration of this complex can successfully reverse drug resistance.
