Drug intelligence / Profile preview
C-terminal alpha-synuclein vaccine is an active immunotherapy candidate developed by Prothena for the treatment and prevention of Parkinson's disease and other synucleinopathies. The vaccine consists of linear tandem C-terminal alpha-synuclein peptides conjugated to the carrier protein CRM197 (a non-toxic mutant of diphtheria toxin) and is formulated with the saponin-based adjuvant QS-21. By targeting the C-terminal region of alpha-synuclein, the vaccine is designed to stimulate the host immune system to produce high-titer antibodies that selectively bind to pathological alpha-synuclein aggregates, block their cellular uptake, and promote their clearance, thereby potentially halting disease progression.
