C-X-C motif chemokine ligand 10 messenger RNA lipid nanoparticle (Cxcl10 mRNA-LNP) is an investigational mRNA-based immunotherapy designed to modulate the tumor microenvironment. It consists of messenger RNA encoding the CXCL10 chemokine (also known as IP-10) encapsulated within a lipid nanoparticle (LNP) delivery system. Upon intramuscular administration, the mRNA is translated into CXCL10 protein, which acts as a potent chemoattractant for activated T cells by signaling through the CXCR3 receptor. This 'genetic adjuvant' approach is intended to increase the infiltration of CD8+ cytotoxic T cells into tumors, particularly in immunotherapy-resistant cancers like melanoma. Preclinical studies have demonstrated that Cxcl10 mRNA-LNP can significantly reduce tumor volume and metastasis when used in combination with immune checkpoint inhibitors such as anti-PD-1 and anti-CTLA-4 antibodies.
