**C027** is a preclinical, replication-competent oncolytic herpes simplex virus type 1 derived from C134 and engineered to express murine interleukin-27. Bicistronic murine IL-27 expression cassettes were inserted into the deleted γ134.5 loci under an MND promoter. C027 is intended for local treatment of malignant glioma, combining direct virus-mediated tumor-cell lysis with intratumoral IL-27 delivery to enhance antitumor T-cell activity, including cytotoxic CD8 T-cell function, T-helper 1 polarization, and durable antitumor immune memory. In syngeneic orthotopic murine glioma models, C027 improved survival; its benefit depended on IL-27 and T cells. ([jitc.bmj.com](https://jitc.bmj.com/content/13/7/e012227))
