Overview

C1142 is a rodent chimeric monoclonal antibody that neutralizes rodent monocyte chemoattractant protein-1 (MCP-1), also known as CCL2 or JE. Developed by Centocor (now Janssen), it serves as a preclinical surrogate for the human anti-CCL2 antibody carlumab (CNTO 888). C1142 is used to evaluate the therapeutic impact of blocking the CCL2/CCR2 signaling axis in rodent models of disease. CCL2 is a potent chemoattractant for monocytes and macrophages; by neutralizing this chemokine, C1142 reduces the infiltration of tumor-associated macrophages and inflammatory cells into tissues. This mechanism has been shown to inhibit tumor growth and angiogenesis in prostate and ovarian cancer models, as well as mitigate vascular remodeling in pulmonary hypertension and preserve blood-brain barrier integrity following intracerebral hemorrhage.
