Overview

**C118P** is a novel small molecule drug that functions as a microtubule inhibitor with **anti-angiogenic** and **vascular disrupting activities**, showing potent anti-tumor effects against solid cancers such as hepatocellular carcinoma and gastric cancer. C118P is a water-soluble analog of combretastatin A4 (CA4) and acts primarily by destabilizing microtubules via binding to the colchicine site on tubulin, leading to cell cycle arrest at G2/M and caspase-dependent apoptosis. Additionally, C118P has been shown to bind directly to RAB1A, promoting its degradation through the autophagy–lysosomal pathway and suppressing mTORC1 signaling, contributing to inhibition of tumor cell proliferation and induction of autophagy. It has demonstrated efficacy in preclinical models, including xenograft mouse studies, and phase I clinical trials have been approved in China for solid tumors including hepatocellular carcinoma and gastric cancer[1][3].
