Overview

C1632 is a small-molecule inhibitor of the RNA-binding proteins Lin28A and Lin28B. Lin28 is an oncofetal protein that is often overexpressed in various cancers, where it functions to suppress the maturation of the let-7 family of microRNAs, thereby promoting pluripotency and cancer stem cell maintenance. C1632 specifically targets the Cold Shock Domain (CSD) of Lin28, interfering with its ability to bind to let-7 precursors. By restoring let-7 levels, C1632 inhibits oncogenic signaling pathways. In preclinical models of hepatocellular carcinoma (HCC), C1632 has been shown to downregulate the expression of immune-suppressive markers such as indoleamine 2,3-dioxygenase-1 (IDO1) and PD-L1, leading to reduced kynurenine production and enhanced efficacy of immunotherapies, including glypican-3 (GPC3)-targeted CAR T-cell therapy.
