Overview

C16G2 is a specifically targeted antimicrobial peptide (STAMP) designed to selectively eliminate *Streptococcus mutans*, the primary bacterial species associated with dental caries. It consists of a 16-amino acid targeting domain (C16) that provides specificity for *S. mutans* and a G2 killing domain derived from the broad-spectrum antimicrobial peptide novispirin G10, joined by three glycine residues. The mechanism of action involves binding to *S. mutans* cell surfaces and disrupting their cytoplasmic membrane, leading to loss of membrane potential, efflux of intracellular contents, and ultimately cell death. This targeted approach allows C16G2 to eradicate *S. mutans* from multispecies oral biofilms without significantly affecting non-cariogenic oral streptococci[1][2][5]. Developed using proprietary STAMP technology by C3 Jian, Inc., C16G2 has completed Phase 2 clinical trials for the prevention of dental caries[1][5][7].
