C18ORF25 siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of the C18ORF25 gene. Research presented at the ADA 2025 meeting indicates that C18ORF25 acts as a regulator of glucose metabolism and an inhibitor of the canonical Wnt/β-catenin signaling pathway in bone tissue. By knocking down C18ORF25, the siRNA promotes metabolic reprogramming—specifically increasing glycolysis and oxidative phosphorylation—which in turn drives osteogenic differentiation of osteoblasts. In preclinical rat models, local intraosseous injection of C18ORF25 siRNA was shown to enhance bone turnover, mass, and strength, suggesting its potential as a therapeutic strategy for metabolic bone diseases such as osteoporosis.
