C225-ILs-dox (anti-EGFR immunoliposomes loaded with doxorubicin) is an experimental targeted nanomedicine developed by the University Hospital Basel for the treatment of relapsed or refractory EGFR-amplified glioblastoma and high-grade gliomas. The drug consists of pegylated liposomal doxorubicin (derived from Caelyx) covalently conjugated to the Fab' fragments of the anti-EGFR monoclonal antibody cetuximab (C225). This design allows for the targeted delivery of the cytotoxic payload, doxorubicin, specifically to tumor cells overexpressing the epidermal growth factor receptor (EGFR), potentially increasing intratumoral drug concentration while minimizing systemic toxicity. It has been evaluated in a Phase 1 pharmacokinetic study known as the GBM-LIPO trial (NCT03603379).
