C2LP-ADC is an experimental antibody-drug conjugate (ADC) developed by ChemPartner, designed to target HER2-positive malignancies such as breast and gastric cancers. The drug utilizes a next-generation "cleavage cascade" linker technology (C2LP) that incorporates two distinct cleavage events. This dual-cleavage mechanism is engineered to enhance linker robustness and reduce the risk of non-specific payload release in systemic circulation, thereby improving the therapeutic index and minimizing off-target toxicity. Preclinical studies presented at AACR 2026 demonstrated that C2LP-ADC possesses superior plasma stability and a better safety profile compared to reference ADCs, with treated subjects maintaining body mass for longer periods. The C2LP platform aims to address common challenges in ADC development, including narrow therapeutic windows and systemic toxicity.
