C34 is a second-generation alpha-galactosylceramide (α-GalCer) analog adjuvant developed by RH Biopharma (RuenHuei Biopharmaceuticals Inc.) for use in therapeutic cancer vaccines. It is specifically designed to enhance the immunogenicity of vaccines targeting the tumor-associated glycolipid antigen Globo H, which is overexpressed in approximately 80% of breast, prostate, and lung cancers. C34 acts as a potent ligand for the CD1d receptor on antigen-presenting cells, such as dendritic cells and B cells. This binding facilitates the presentation of the glycolipid to invariant natural killer T (iNKT) cells, triggering their activation and the subsequent secretion of cytokines that promote a robust adaptive immune response. In preclinical studies, C34-adjuvanted vaccines have demonstrated the ability to induce high-affinity IgG antibodies against Globo H and related antigens SSEA-3 and SSEA-4. The drug is currently in preclinical development, with ongoing efforts focused on optimizing its water-soluble formulation and industrial manufacturing processes.
