c8-glucosylceramide (also known as C8-GluCer or N-octanoyl-glucosylceramide) is a synthetic short-chain sphingolipid and glycosphingolipid analog. It is primarily investigated as a membrane-modulating adjuvant to enhance the intracellular delivery and therapeutic efficacy of chemotherapeutic agents, such as doxorubicin and mitoxantrone. By inserting into the tumor cell plasma membrane, c8-glucosylceramide increases membrane permeability and facilitates the transmembrane traversal of amphiphilic drugs, thereby overcoming barriers to drug uptake and reversing multidrug resistance. Preclinical research, primarily led by institutions like the Netherlands Cancer Institute and Erasmus University Medical Center, has explored its co-formulation within liposomes or as a pretreatment strategy to improve chemotherapy outcomes in solid tumors, including melanoma and breast carcinoma.
