C9 single-chain variable fragment is a recombinant, human-derived antibody fragment developed by Var2 Pharmaceuticals that specifically targets oncofetal chondroitin sulfate (ofCS). ofCS is a unique glycosaminoglycan modification that is essentially absent from healthy adult tissues but is highly expressed on the surface of a wide variety of cancer cells and in the placenta. The C9 scFv was discovered through phage display screening to identify binders that mimic the high-affinity interaction between the malaria protein VAR2CSA and ofCS. This scFv is utilized as a modular targeting domain in the development of next-generation cancer therapies, including Chimeric Antigen Receptor (CAR) T-cells and bispecific antibodies, aimed at treating a broad spectrum of solid tumors such as colorectal, pancreatic, and lung carcinomas.
