C-93 is a potent, synthetic small molecule inhibitor of Fatty Acid Synthase (FASN), developed as a successor to the earlier compound C75. FASN is a multi-enzyme complex that catalyzes the de novo synthesis of long-chain fatty acids, a process that is significantly upregulated in many cancers to support rapid membrane synthesis and signaling. C-93 works by inhibiting the ketoacyl synthase (KS) domain of FASN, leading to the accumulation of malonyl-CoA. This biochemical shift not only induces apoptosis in cancer cells (particularly in lung and prostate cancers) but also suppresses appetite and increases fatty acid oxidation by inhibiting carnitine palmitoyltransferase-1 (CPT-1). Developed by FASgen in collaboration with Johns Hopkins University, C-93 was designed to minimize the transient stimulation of fatty acid oxidation seen with C75, thereby potentially offering a better therapeutic window for treating obesity and cancer.
