CA028 is a synthetic isoflavone derivative of calycosin (7-hydroxy-3-(3-hydroxy-4-methoxyphenyl) chromen-4-one) developed for the treatment of various cancers, including melanoma, oral cancer, and colorectal cancer. Chemically, it is modified from calycosin by the substitution of two hydrogen atoms with ethyl acetate groups (CH2COOCH2CH3), resulting in the molecular formula C24H24O9. CA028 exerts anti-tumor effects by inhibiting cell proliferation, migration, and invasion while inducing apoptosis. Its mechanism of action involves the modulation of multiple signaling pathways, most notably the p53 and death receptor signaling pathways, and the induction of genes such as BAK1, CDKN1A (p21), SUFU, and BMP4. Preclinical studies in melanoma models have demonstrated significant tumor growth inhibition in vivo and reduced metastatic potential in vitro.
