Overview

CA102L is a polymer-drug conjugate composed of the polysaccharide hyaluronic acid (HA) covalently linked to lenalidomide, an immunomodulatory drug (IMiD). Developed by Holy Stone Healthcare, CA102L utilizes the high affinity of HA for the CD44 receptor, which is frequently overexpressed in multiple myeloma (MM) and associated with disease progression. This targeting mechanism is intended to increase the local concentration of lenalidomide within tumor cells while minimizing systemic off-target toxicity. Upon internalization, the lenalidomide payload interacts with its primary molecular target, cereblon (CRBN), a component of the CRL4-CRBN E3 ubiquitin ligase complex. This interaction leads to the ubiquitination and subsequent proteasomal degradation of transcription factors such as Ikaros (IKZF1) and Aiolos (IKZF3), resulting in potent antiproliferative and antiangiogenic effects in MM models.
