CA9-GM-CSF fusion protein is an experimental immunotherapeutic agent designed to treat Carbonic Anhydrase IX (CA9) expressing malignancies, most notably clear cell renal cell carcinoma (ccRCC). The molecule consists of the tumor-associated antigen CA9 fused to the cytokine granulocyte-macrophage colony-stimulating factor (GM-CSF). This dual-function construct aims to direct the immune system toward tumor cells expressing CA9 while simultaneously utilizing GM-CSF to recruit and activate antigen-presenting cells (APCs), such as dendritic cells, thereby enhancing the priming of tumor-specific T-cells. It has been investigated primarily as a DNA vaccine (plasmid DNA encoding the fusion protein) and in cell-based therapies where dendritic cells are transfected with the fusion construct. The approach was developed at the University of California, Los Angeles (UCLA) and is covered under patents describing novel CA9 variants (CAIXv) for enhanced immunogenicity.
