Overview

CA9hu-2 is a humanized monoclonal antibody developed by Mabpro that targets Carbonic Anhydrase IX (CA IX), a protein highly expressed in the hypoxic microenvironment of various solid tumors. As the humanized version of the murine antibody IV/18, CA9hu-2 specifically binds to the sequential epitope of the proteoglycan domain of CA IX. Unlike its counterpart CA9hu-1, CA9hu-2 does not internalize upon binding, which enables it to effectively block the attachment of cancer cells to the extracellular matrix, thereby inhibiting the formation of metastases. Additionally, the antibody triggers potent immune-mediated tumor cell killing through antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cell-mediated phagocytosis (ADPC), and complement-dependent cytotoxicity (CDC). It is currently in preclinical development for the treatment of triple-negative breast cancer (TNBC) and clear cell renal cell carcinoma (ccRCC).
