Overview

CAB-B7-H3 is a conditionally active bispecific monoclonal antibody being developed by Himalaya Therapeutics. Designed as a T-cell engager, it simultaneously targets the B7-H3 (CD276) antigen on tumor cells and the CD3 receptor on T cells. The "CAB" (Conditionally Active Biologic) technology allows the antibody to bind preferentially to its targets within the acidic microenvironment characteristic of tumors, which is intended to minimize off-target binding and toxicity in healthy tissues. By recruiting and activating T cells in close proximity to B7-H3-expressing cancer cells, the drug facilitates direct T-cell mediated cytotoxicity. It is currently under investigation for the treatment of various solid tumors, including head and neck cancer, melanoma, lung cancers, and several other refractory malignancies.
