Overview

CAB-EGFR is a conditionally active bispecific (CAB) antibody developed by Himalaya Therapeutics, utilizing technology designed to selectively target the tumor microenvironment. As a bispecific T-cell engager, it simultaneously binds to the Epidermal Growth Factor Receptor (EGFR) on malignant cells and the CD3 subunit on T cells. This dual engagement facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent directed lysis of EGFR-expressing tumor cells. The 'conditionally active' nature of the molecule is intended to restrict its activity to the unique conditions of the tumor microenvironment (such as acidic pH), thereby minimizing 'on-target, off-tumor' toxicities typically associated with EGFR-targeted therapies in healthy tissues like the skin and gut. It is currently being investigated for the treatment of solid tumors, including bladder cancer, pancreatic cancer, and triple-negative breast cancer (TNBC).
