Overview

CAB-Nectin-4 is a conditionally active bispecific antibody (CAB) being developed by Himalaya Therapeutics for the treatment of multiple solid tumor types. Utilizing Conditionally Active Biologic (CAB) technology, the antibody is designed to bind its targets preferentially within the unique conditions of the tumor microenvironment (such as low pH), which aims to minimize off-target binding to healthy tissues. The molecule functions as a T-cell engager, simultaneously targeting Nectin-4 (a cell surface protein overexpressed in various carcinomas including bladder, breast, and lung cancers) and the CD3 receptor on T cells. This dual binding facilitates the recruitment and activation of cytotoxic T cells directly to the tumor site, leading to the targeted destruction of Nectin-4-expressing malignant cells.
