CAB102 is a humanized monoclonal antibody designed to target the growth factor midkine (MK). Originally developed by the Australian biotechnology company Cellmid (now Anagenics) in collaboration with Antitope Ltd, the antibody was humanized using the proprietary Composite Human Antibody™ technology to minimize immunogenicity. CAB102 specifically targets the C-domain of midkine, a region essential for its pathological signaling in various diseases. Midkine is a heparin-binding growth factor that is overexpressed in many cancers and inflammatory conditions, where it promotes cell survival, migration, and inflammation. The therapeutic program was managed by Cellmid's subsidiary Lyramid and was acquired by Roquefort Therapeutics in 2021. Preclinical studies investigated CAB102 for multiple indications, including solid tumors (to overcome chemotherapy resistance), focal segmental glomerulosclerosis (FSGS), chronic kidney disease, multiple sclerosis, rheumatoid arthritis, and myocarditis. Despite showing promise in preclinical models, the drug did not advance to human clinical trials.
