Cabazitaxel + bavituximab is an investigational combination therapy for cancer, primarily studied in metastatic castration-resistant prostate cancer. Cabazitaxel is a second-generation, semisynthetic taxane microtubule inhibitor that stabilizes microtubules by binding to the beta-tubulin subunit, thereby preventing cell division and inducing tumor cell death. It is less susceptible to P-glycoprotein-mediated drug resistance and can cross the blood–brain barrier[2][5][8]. Bavituximab is a chimeric IgG1 monoclonal antibody that acts as a vascular disrupting agent by targeting phosphatidylserine (PS), an anionic phospholipid externalized on tumor blood vessels under stress conditions such as hypoxia or acidity. By binding PS in complex with beta 2 glycoprotein 1, bavituximab selectively disrupts tumor vasculature[6]. The combination has been evaluated in early-phase clinical trials for advanced cancers[3].
