Overview
Cabazitaxel liposome is a liposomal formulation of the semi-synthetic taxane cabazitaxel, developed to improve the therapeutic index of the drug. Cabazitaxel exerts its antineoplastic effect by binding to and stabilizing tubulin, which inhibits microtubule depolymerization, leading to G2/M phase cell cycle arrest and apoptosis. The liposomal delivery system utilizes a lipid bilayer to encapsulate the drug, which facilitates preferential accumulation in tumor tissues through the enhanced permeability and retention (EPR) effect. This formulation is specifically designed to eliminate the need for surfactants like polysorbate 80 (Tween 80), which are used in the standard formulation (Jevtana) and are associated with severe hypersensitivity reactions and the requirement for corticosteroid premedication. Currently, it is being investigated in Phase 1 clinical trials by developers such as Lunan Pharmaceutical and Shanghai Pharma for the treatment of advanced solid tumors and metastatic castration-resistant prostate cancer.
