Overview

cabergoline + hydroxychloroquine is an experimental combination therapy being evaluated for the treatment of cabergoline-resistant prolactinomas. Cabergoline is a potent dopamine D2 receptor agonist that serves as the standard of care for prolactin-secreting pituitary adenomas by inhibiting prolactin secretion and inducing tumor shrinkage. In cases of resistance, tumor cells often utilize autophagy as a survival mechanism to evade apoptosis. Hydroxychloroquine, a 4-aminoquinoline derivative, acts as an autophagy inhibitor by increasing lysosomal pH and preventing autophagosome-lysosome fusion. By combining these two agents, the treatment aims to sensitize resistant prolactinoma cells to the growth-inhibitory effects of cabergoline. This approach is currently being studied in clinical trials led by investigator Zhebao Wu.
