Cabiralizumab is a humanized monoclonal antibody that targets the colony-stimulating factor 1 receptor (CSF1R), also known as macrophage colony-stimulating factor receptor (M-CSFR) and CD115. By binding to CSF1R on monocytes, macrophages, and osteoclasts, it competitively inhibits the binding of its ligands—colony stimulating factor 1 (CSF1) and interleukin-34 (IL-34)—thereby blocking downstream signaling. This action reduces the survival and function of tumor-associated macrophages (TAMs), which are implicated in immune suppression within tumors, tumor growth, angiogenesis, invasion, metastasis, and drug resistance. Cabiralizumab has been investigated for various cancers including pancreatic cancer, tenosynovial giant cell tumor/pigmented villonodular synovitis (TGCT/PVNS), malignant melanoma, non-small cell lung cancer (NSCLC), renal cell carcinoma (RCC), solid tumors, triple negative breast cancer as well as immune-mediated diseases such as rheumatoid arthritis. It was originally developed by Five Prime Therapeutics and later co-developed with Bristol Myers Squibb; Amgen acquired Five Prime Therapeutics in 2021[4][5][6].
