Overview

Cabotamig (ARB202) is a first-in-class bispecific T-cell engager (BiTE) antibody designed to target Cadherin-17 (CDH17) and the CD3 receptor on T cells. Developed by Arbele, the drug is specifically engineered to treat advanced gastrointestinal (GI) malignancies, including colorectal, gastric, and pancreatic cancers, where CDH17 is highly and specifically expressed. The mechanism of action involves the simultaneous binding of CDH17 on the surface of tumor cells and the CD3 epsilon subunit of the T-cell receptor complex. This dual binding facilitates the formation of an immunological synapse, leading to the activation of T cells and the subsequent release of perforins and granzymes, which results in the targeted lysis of CDH17-positive cancer cells. Cabotamig is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and preliminary efficacy in patients with CDH17-positive advanced solid tumors who have failed standard therapies.
