A combination therapy consisting of cabozantinib, a multi-tyrosine kinase inhibitor targeting VEGFR2, MET, AXL, RET, KIT, FLT3, and TIE-2, and temozolomide, an alkylating agent that damages DNA in cancer cells. This combination has demonstrated synergistic anticancer effects in preclinical studies and clinical trials, particularly for soft tissue sarcomas including leiomyosarcoma. The dual targeting of VEGF and c-MET pathways with cabozantinib, combined with temozolomide's DNA-damaging effects, produces a synergistic antitumor effect.
