CAd12_PDL1 is a binary oncolytic adenovirus (CAd) designed to enhance the efficacy of chimeric antigen receptor (CAR) T-cell therapies in solid tumors. It functions by inducing local oncolysis and expressing two therapeutic payloads: the cytokine interleukin-12 (IL-12p70) and a programmed death-ligand 1 (PD-L1) blocking antibody. The IL-12 component is intended to prevent the loss of CAR T cells at the tumor site and promote their persistence, while the anti-PD-L1 antibody helps overcome the immunosuppressive tumor microenvironment. In preclinical models of head and neck squamous cell carcinoma (HNSCC), CAd12_PDL1 has demonstrated the ability to augment the anti-tumor effects of HER2-specific CAR T cells, leading to improved survival and control of both primary and metastatic tumors.
