cAd3-EBOV is a replication-deficient chimpanzee adenovirus serotype 3 (cAd3) viral vector vaccine developed to prevent Ebola virus disease. Developed by GSK in collaboration with the National Institute of Allergy and Infectious Diseases (NIAID), the vaccine utilizes the cAd3 vector to deliver the gene encoding the surface glycoprotein (GP) of the Ebola virus. Upon intramuscular administration, the vector enters host cells and expresses the GP antigen, which triggers both humoral and cellular immune responses. The vaccine has been evaluated in several formulations, including monovalent versions targeting the Zaire ebolavirus (cAd3-EBOZ) or Sudan ebolavirus (cAd3-EBO S), and a bivalent version (cAd3-EBO) targeting both. While clinical trials demonstrated that the vaccine is safe and immunogenic, its development was not pursued to regulatory approval as other candidates were prioritized during the 2014-2016 West African outbreak.
