Drug intelligence / Profile preview
CADM1-directed CAR-T cells are chimeric antigen receptor (CAR) T-cell therapies engineered to target Cell Adhesion Molecule 1 (CADM1), a cell-surface glycoprotein overexpressed in neuroendocrine tumors, small cell lung cancer (SCLC), and lung adenocarcinoma (LUAD). Developed by researchers at the University of Michigan, these CAR-T cells recognize CADM1 on tumor cells, leading to antigen-specific activation, secretion of pro-inflammatory cytokines (such as IFN-γ and TNF-α), and upregulation of cytolytic molecules (granzymes and perforin) to induce tumor cell lysis. Preclinical studies have demonstrated robust anti-tumor efficacy and inhibition of metastasis in SCLC and LUAD models, with minimal on-target/off-tumor toxicity against normal tissues.
