AK104 + axitinib is an investigational combination therapy comprising cadonilimab (AK104), a bispecific antibody targeting PD-1 and CTLA-4, and axitinib, a small molecule tyrosine kinase inhibitor of VEGFR-1, VEGFR-2, and VEGFR-3. This combination is designed to provide dual immune checkpoint blockade while simultaneously inhibiting tumor angiogenesis. It is currently being evaluated in Phase Ib clinical trials, specifically for the treatment of metastatic mucosal melanoma, a rare and aggressive subtype of melanoma. Cadonilimab is developed by Akeso, while axitinib is a multi-target TKI originally developed by Pfizer.
