This combination therapy involves the bispecific antibody cadonilimab (AK104) administered alongside standard-of-care chemotherapy regimens, specifically Gemcitabine plus Cisplatin (GP) or a regimen containing 5-Fluorouracil and Cisplatin (PFLL). Cadonilimab is a first-in-class tetravalent bispecific antibody that simultaneously targets the programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) immune checkpoints. By blocking both pathways, it aims to enhance anti-tumor immune responses more effectively than single-checkpoint inhibitors. The chemotherapy components (gemcitabine, cisplatin, and 5-fluorouracil) provide cytotoxic effects by inhibiting DNA synthesis and inducing DNA damage. This specific combination is being evaluated by Sun Yat-sen University Cancer Center for the treatment of patients with recurrent or metastatic nasopharyngeal carcinoma who have failed first-line therapy.
