Overview

Cadonilimab + SOX is a combination therapy consisting of **cadonilimab**, a tetrameric bispecific antibody targeting both programmed death-1 (**PD-1**) and cytotoxic T-lymphocyte-associated protein 4 (**CTLA-4**), together with the **SOX chemotherapy regimen**. The SOX regimen includes **S-1** (an oral fluoropyrimidine derivative composed of tegafur, gimeracil, and oteracil potassium) and **oxaliplatin** (a platinum-based chemotherapy agent). Cadonilimab acts as a dual immune checkpoint inhibitor, aiming to enhance anti-tumor immune responses. The combination is primarily in development for the neoadjuvant and first-line treatment of **locally advanced and advanced/metastatic gastric or gastroesophageal junction adenocarcinoma**. Studies suggest that this combination may improve pathological complete response and R0 resection rates compared to standard therapy, offering a potentially more effective neoadjuvant approach[1][3][5][8]. Cadonilimab is developed by Akeso Biopharma and is approved in China for recurrent or metastatic cervical cancer.
