CAdTetra is a binary oncolytic/helper-dependent adenovirus system (CAdVEC) developed by researchers at the Baylor College of Medicine. It is designed to enhance local and systemic antitumor immunity against solid tumors, including cancer stem cell populations. CAdTetra is built upon the CAdTrio platform, which co-expresses interleukin-12 (IL-12), a programmed death-ligand 1 (PD-L1) blocker, and a herpes simplex virus thymidine kinase (HSV-tk) safety switch. CAdTetra additionally incorporates a bispecific T-cell engager (BiTE) targeting CD44v6. This design redirects tumor-infiltrating, virus-specific (or other irrelevant) T cells to target and eliminate CD44v6-expressing cancer stem cells, thereby overcoming the immunosuppressive tumor microenvironment and inducing robust adaptive antitumor immune responses.
