Overview

CAdTrio is an investigational **binary oncolytic/helper-dependent adenoviral immunotherapy** developed at Baylor College of Medicine. It comprises a replication-competent oncolytic adenovirus and a non-replicating helper-dependent adenovirus engineered for local intratumoral delivery of interleukin-12, a programmed cell death ligand 1 blocking mini-antibody, and an HSV thymidine kinase safety switch. The oncolytic component lyses infected tumor cells, while local IL-12 expression promotes antitumor immune activation and PD-L1 blockade reduces tumor-mediated immune suppression. CAdTrio has been evaluated clinically as the CAdVEC system in a Phase 1 study of advanced or refractory HER2-positive solid tumors, including breast cancer.
