Overview

CAF10b is a novel liposomal vaccine adjuvant designed to stimulate robust Th1 and Th17 immune responses, particularly memory antibody and T cell responses. It incorporates a Toll-like receptor 9 (TLR9) agonist to enhance immunogenicity. CAF10b is being evaluated as part of the H107e/CAF10b tuberculosis (TB) subunit vaccine, which combines eight Mycobacterium tuberculosis antigens with this adjuvant. The formulation aims to complement BCG vaccination by inducing strong cellular immunity against TB-specific antigens and has demonstrated significant enhancement of both antibody and cellular immune responses in non-human primates compared to previous adjuvants. Its primary route of administration is intramuscular injection, and it is currently in phase 1 clinical trials for TB prevention[1][2][3][4][5][6].
