Overview

caIL-12 OT-I TCR-T cells are an experimental adoptive T-cell therapy consisting of T cells engineered to express both a specific T-cell receptor (OT-I) and a membrane-anchored, constitutively active form of interleukin-12 (caIL-12). The OT-I TCR specifically recognizes the SIINFEKL peptide of the ovalbumin (OVA) protein. The inclusion of membrane-restricted IL-12 is designed to provide localized cytokine support within the tumor microenvironment, enhancing the potency of the T cells and promoting antigen spreading—the activation of endogenous T cells against other tumor antigens—thereby overcoming tumor heterogeneity and antigen escape. This approach aims to achieve the potent immunostimulatory effects of IL-12 while avoiding the severe systemic toxicity associated with its recombinant or secreted forms.
