Overview

Calcein, also known as Oftasceine or Fluorexon, is a fluorescent dye belonging to the xanthene family. Its acetoxymethyl ester derivative, Calcein AM, is a non-fluorescent, cell-permeant compound. Upon entering live cells, intracellular esterases hydrolyze Calcein AM, converting it into highly fluorescent Calcein, which is then retained within the cell. This mechanism makes Calcein a valuable tool for assessing cell viability, membrane integrity, and for short-term cell labeling in various biological assays. It is also used as an indicator for lipid vesicle leakage and as an ophthalmic staining agent in procedures like Goldmann applanation tonometry and contact lens fitting. Calcein's fluorescence properties allow for the visualization and quantification of cellular processes and structures.
