Calcipotriene is a synthetic small molecule and vitamin D3 analogue used primarily for the topical treatment of chronic plaque psoriasis. It acts as an agonist at the vitamin D receptor (VDR), which is a nuclear transcription factor that regulates gene expression involved in skin cell growth and immunity. By binding to VDR, calcipotriene inhibits the abnormal overproduction and proliferation of keratinocytes and induces their differentiation, thereby normalizing epidermal growth and reducing the redness, scaling, and thickness of psoriasis plaques. It is commonly formulated as an ointment, cream, foam, or scalp solution and is often used in combination with corticosteroids like betamethasone dipropionate to enhance efficacy and reduce irritation.
