Calcipotriol is a synthetic derivative of calcitriol (active vitamin D3) used topically to treat plaque psoriasis. It works by binding to the vitamin D receptor (VDR) on epidermal and immune cells, modulating gene expression involved in cell differentiation and proliferation. This leads to reduced excessive skin cell production characteristic of psoriasis plaques. Calcipotriol has much lower activity in calcium metabolism than natural calcitriol, making it suitable for topical use without significant systemic effects. The drug is available as ointment, cream, or scalp solution and is sometimes combined with corticosteroids for enhanced efficacy[1][2][3][4][5][6].
