This is a combination regimen of three drugs—calcitriol (the active form of vitamin D3), cisplatin (a platinum-based chemotherapy agent), and docetaxel (a taxane-class chemotherapy drug). Calcitriol acts as a hormone with antiproliferative effects on cancer cells, modulating cell growth, differentiation, and apoptosis. Cisplatin forms DNA crosslinks that inhibit DNA synthesis and function, leading to cell death. Docetaxel stabilizes microtubules to prevent cell division. Preclinical studies have shown synergistic antiproliferative effects when calcitriol is combined with cisplatin or docetaxel. This combination has been investigated in phase I/II clinical trials for advanced non-small-cell lung cancer (NSCLC) to determine safety, dosing, pharmacokinetics/pharmacogenomics correlations (notably CYP24A1 SNPs), and preliminary efficacy[1]. The regimen has also been explored in prostate cancer settings for its potential to enhance antitumor activity[3][4][5].
