Calcium and magnesium infusions are used as a supportive care intervention to prevent or mitigate oxaliplatin-induced peripheral sensory neuropathy in patients with colorectal cancer. Oxaliplatin, a platinum-based chemotherapy agent, produces oxalate as a metabolite, which is thought to chelate intracellular calcium and magnesium, thereby altering the function of voltage-gated sodium channels in sensory neurons. By providing exogenous calcium and magnesium intravenously before and after oxaliplatin administration, clinicians aim to stabilize these channels and reduce neurotoxicity. In the Phase III GLUTOX trial (NCT01087658) sponsored by Sanofi, this combination served as the control arm against which the addition of oral glutamine was evaluated to assess its efficacy in reducing grade 2-4 peripheral sensory neuropathy.
