CALY-001

CALY-001 is a fully human therapeutic monoclonal antibody that selectively binds to and neutralizes the active form of matrix metalloproteinase 9 (MMP-9), a zinc-dependent endopeptidase that degrades extracellular matrix and contributes to inflammation, tissue damage, and fibrogenesis in the gut and other organs.[2][3][5][7][13] By targeting only active MMP-9 and sparing the abundant pro-enzyme, CALY-001 is designed to maintain full inhibition of pathogenic MMP-9 activity while avoiding target-mediated clearance, potentially enabling more sustained exposure and improved efficacy versus earlier anti–MMP-9 approaches.[2][4][5][7] Originally derived from research at Merck and spun out into Calypso Biotech, CALY-001 has been globally licensed to EA Pharma for development in inflammatory bowel disease and is also being positioned preclinically for additional oncology, inflammatory, and fibrotic indications.[2][3][4][11][12][13]
